General description

Superoxide Dismutase from bovine erythrocytes is a metalloprotein which disproportionates superoxide anion radicals.[1] It is a 31.5 kDa copper binding protein and displays a conserved domain and fold.[2] It is a homodimer with one copper and zinc ion per subunit and has antiparallel “greek-key” β barrel fold.[3]

Application

Superoxide Dismutase (SOD) from bovine erythrocytes has been used:
• for measuring the superoxide radical using the electron paramagnetic resonance spin in human brain microvascular endothelial cells[4]
• for measuring superoxide production in cytochrome C assay in peripheral blood mononuclear cells[5]
• as a standard in characterization of hen egg SOD using Fourier-transform infrared spectroscopy (FTIR) and matrix-assisted laser desorption/ionization (MALDI) analysis[6]

Biochem/physiol Actions

催化超氧化物自由基的歧化反应生成过氧化氢和分子氧。在细胞防御氧自由基的毒性作用方面发挥着重要作用。与一氧化氮(NO)竞争超氧化物阴离子（可与NO反应生成过亚硝酸盐），因此SOD能促进NO的活性。在培养的大鼠卵泡、神经细胞系和转基因小鼠中，SOD也表现出抑制细胞凋亡的作用。

Unit Definition

在pH 7.8，25℃条件下，3.0mL反应体积中，一个单位可在黄嘌呤氧化酶的偶联系统中抑制细胞色素c的减少达到50％。黄嘌呤氧化酶浓度应产生初始ΔA550=0.025±0.005/分钟。
